Benchmark Accuris SmartGlow DNA Stains are ideal for excitation of nucleic acids with UV or visible blue light transilluminators. Unlike Ethidium Bromide (EtBR), Benchmark Accuris SmartGlow DNA Stains are non-mutagenic and do not require special methods of disposal.
The Benchmark Accuris SmartGlow DNA Stains - SmartGlow PS (Pre Stain) can be used as a direct replacement for EtBR in agarose and polyacrylamide gel electrophoresis. The Benchmark Accuris SmartGlow DNA Stains - SmartGlow PS (Pre Stain) emits green fluorescence when bound to dsDNA or ssDNA and emits red fluorescence when bound to RNA. The SmartGlow PS (Pre Stain) exhibits excitation peaks at 290 nm and 490 nm, allowing it to be used with both UV and blue light.
The Benchmark Accuris SmartGlow DNA Stains - SmartGlow LD (Loading Dye) is supplied at a 6X concentration and is added directly to your samples. No dye then needs to be added to the gel or running buffer. After electrophoretic separation, you can simply view and document your results using a UV or blue light illuminator. The Benchmark Accuris SmartGlow DNA Stains - SmartGlow LD (Loading Dye) emits green fluorescence when bound to dsDNA, ssDNA and RNA.

Protocol for SmartGlow PS

Prepare 100 mL of agarose or polyacrylamide solution.

Add 5 µl of SmartGlow PS stain to the gel solution before pouring gels.

For enhanced results, add SmartGlow PS to the running buffer at a ratio of 2.5 µl to 5 µl per 100 mL. Adding SmartGlow PS to the running buffer will result in increased sensitivity & better detection of small quantities of nucleic acid.

After electrophoresis is complete, view the gel using a UV or blue light illuminator

Prepare agarose for polyacrylamide solution.

Pour gels.

Mix SmartGlow LD with samples & DNA markers at a 1:5 (dye to sample) dilution.
